چکیده
background: herbal medicine is a new complementary strategy for treating diseases such as cancer. however, its mechanism of action is not clear yet. this study investigates antitumor efficacy of polygonum avicular extract on mcf-7 cell line . methods: mcf-7 cells were treated with different concentrations of (50, 100, 150, 200, 250, 300,350 400 ng/âµl) polygonum avicular at different time intervals (6, 12, 24, and 48 hrs). in order to evaluate the expression level of fasl and bcl2 genes, rt-pcr technique was employed. results: the obtained results demonstrate that expression of fasl gene was increased in polygonum avicular treated cells in comparison to control group. down-regulation of bcl2 expression was observed during the treatment. conclusion: polygonum aviculare herbal extract may induce apoptosis through up-regulation of fasl and down-regulation of bcl2 gene, however, its complete mechanism of action must be elucidated.

Bronchodilatory and anticholinergic effects of Polygonum avicular on isolated tracheal chains of the guinea pig
In this study we investigated the bronchodilatory and anticholinergic effects of the aqueous extract (AE), brewed extract (BE) and ethanol extract (EE) of Polygonum avicular in comparison with saline (S) on isolated tracheal chains of the guinea pig. Due to the probable confounding effect of ethanol in the EE, the effects of this extract were also compared with 96 % ethanol. None of the plant...
متن کامل5-azacytidine and decitabine exert proapoptotic effects on neoplastic mast cells: role of FAS-demethylation and FAS re-expression, and synergism with FAS-ligand.
Aggressive systemic mastocytosis (ASM) and mast cell leukemia (MCL) are advanced hematopoietic neoplasms with poor prognosis. In these patients, neoplastic mast cells (MCs) are resistant against various drugs. We examined the effects of 2 demethylating agents, 5-azacytidine and decitabine on growth and survival of neoplastic MCs and the MC line HMC-1. Two HMC-1 subclones were used, HMC-1.1 lack...
متن کاملStatus of FAS and FAS Ligand Gene Polymorphisms in Patients wi th Breast Cancer in Northeastern IRAN
Background: The First apoptosis signal (FAS) and First apoptosis signal ligand (FASL) genes initiate the apoptosis pathway, playing a central role in the tumor growth and metastasis. Gene polymorphisms including -1377 G/A in the promoter region of FAS and -844 C/T in the promoter region of FASL have shown to change the transcription activities of these genes. Methods: In this study we evalua...
متن کاملFas/Fas ligand pathway
The orphan nuclear receptor Nur77/N10 has recently been demonstrated to be involved in apoptosis of T cell hybridomas. We report here that chronic expression of Nur77/N10 in thymocytes of transgenic mice results in a dramatic reduction of CD4+CD8+ double-positive as well as CD4+CD8and CD4-CD8+ single-positive cell populations due to an early onset ofapoptosis. CD4-CD8double-negative and CD25+ p...
متن کاملAqueous Humor Levels of Soluble Fas and Fas-ligand in Patients with Primary Open Angle and Pseudoex-foliation Glaucoma
Background: Glaucoma is one of the most common causes of blindness and is usually associated with elevated intraocular pressure. In patients with primary open angle glau-coma the number of trabecular meshwork cells is decreased. Death of the trabecular meshwork cells may be a result of apoptosis. Objective: To investigate the aqueous humor levels of soluble Fas (sFas) and Fas-Ligand (sFasL) in ...
متن کاملthe role of fas/fas ligand system in the pathogenesis of liver cirrhosis and hepatocellular carcinoma
background the fas receptor/ligand system including soluble forms is the most important apoptotic initiator in the liver. dysregulation of this pathway may contribute to abnormal cell proliferation and cell death and is regarded as one of the mechanisms preventing the immune system from rejecting the tumor cells. objectives to analyze the role of fas system fas/ fas ligand (fas/ fasl) in the mu...
